Well, my bacteria from my ligation did not grow, again. So something is still not quite right in this experiment. It's frustrating because, much like the PCR issues of last month, this should be a piece of cake.
I also tried to do a midi-prep (extracting DNA from other bacterial samples). This was going just fine until the high speed centrifuge broke my tubes and spilled my sample everywhere. Then I had to stop the experiment and will have to start all over again.
I am so frustrated that none of the benchwork seems to be working. I'm doing things from actual kits, you know, the kind of stuff that is supposed to be idiot-proof. In the past, I have had no problems, but this week, it seems as though the gods of biological science are fighting against me.
On an up side, I did locate two different sequences that would be appropriate to use for a Southern blot. Now if I could just get to the place where I would actually need those...
